ETA: for your night time activity. Since everyone else will recommend day stuff. West End theater is wonderful, but you can only see the Globe at the Globe. Obviously you should go to the theater many nights, though.

If I had never been there, Tower of London with tickets for the Ceremony of the Keys that night.
Since I've been there, and done several things (including a show, just not at the Globe), I would want to go mudlarking on the Thames. We were never able to get down when the water was low, but it's something I want to do someday.
I could spend all day at the Tower of London, but you could "fit it in" in four hours or so. I would think that would give enough time for Westminster (but we only walked by that, didn't go in).
We're thinking Tower of London and Westminster. How much time would you say is enough at each to feel like you got a good visit in?
OH. And how the hell do people get from Heathrow to central London without spending a million dollars!?!
As pps mentioned, you can take the Tube from Heathrow to central London on the Picadilly line. If you don't fancy that, you can take the Heathrow Express for around £15 each, and it will take you to Paddington station. A cab will cost you around £50.
Keep in mind that the Abbey usually has a massively long queue to enter during tourist season, so I'd probably go there first, nice and early. It's takes about 1-2 hours to walk around inside without a guided tour. The Tower will probably take you a few hours to explore. The Eye is overpriced and overrated, but a walk along the south bank will give you impressive views of parliament and St Paul's. Once you've finished at the Abbey, cross the Westminster Bridge and head east along the south bank towards Tower Bridge. On the way you can stop to eat lunch at Borough Market (open Tues-Sat). Cheap and amazing food. For dinner, you can't go wrong with a proper pub or a nice curry.
