I'm not sure if it matters. I think I read once not to use FMU, though. Not sure if that's accurate or not though. I've always done them in the late morning, but that's just my preference. I usually just do once, but sometimes twice if I think it's a day I'll really get a positive.

i always did FMU and then again in the afternoon/evening, about 6pm. the only reason they say not to use FMU is that the hormones it detects don't "build up" the way pregnancy hormones do, so it's not like you're better off doing it first thing....but you're not worse off either. you're testing for your surge, which is why they recommend testing twice daily. if you have a short surge in the afternoon, it wouldn't have happened in the morning and might be gone by the next morning. so you'd never get a positive even though your hormones are doing their thing.
and if you REALLY want to know if you're O-ing, you gotta temp/chart too. OPKs won't tell you that.
I test FMU and then when the line starts to darker I test around 5/6pm. I usually get my peak in the evening/early morning. (When I work noon-midnight I test when I get home at midnight as it's easier.)

I use the CBE digital OPKs. I start testing at CD10, and when there is a flashing smiley, I test twice. I test first thing in the morning, then again before I go to bed (if I'm testing twice that day). I normally have a longer surge, so before we started IUIs, I'd only test once, in the morning.
I used to do when I got home from work. My OB wants me to test in the morning, and no way am I doing it at work, so FMU it is. I personally usually have a 2 day surge so it doesn't really make a huge difference to me.
I usually do once a day in the morning, starting around CD 10. Then twice, once in the morning and once in the evening, when when the line gets darker - usually around CD 13-15.
FWIW, I use FMU and most of the time the line is the same when compared to the evening test.
