For quick/cheap meals, I like Fast & French. For something nicer, 82 Queen. Their shrimp and grits are the best I've had in Charleston. Caviar & Bananas has amazing cupcakes.
My favorite thing to do is walk around the historic district and people watch. There is a great park for people-watching at the corner of King St and Calhoun St.
Also, it sounds cheesy but I really enjoy the carriage tours. You always get something different depending on the guide. Last time we were there, we noticed that Sunday morning seems like a really good time to go to avoid being packed into a carriage. We were walking around and it seemed like the tours had at most 4 people per carriage.
I also like to shop on King St. since my town's mall is ridiculously small.
Agree with PP about the things to do as well as taking the ferry and touring Ft. Sumpter, Take a plantation tour. Middleton Place is my absolute favorite. The gardens & the view are gorgeous.
We were there for one day in May and had a lovely time. We had lunch at Husk which I would highly recommend (you will probably need to make reservations in advance), took a carriage tour, went through one of the historic mansions, and just walked around town.
