savannah has a few different ghost tours that are interesting if you like that stuff. also, i loved "the Midnight in the Garden of Good and Evil" tour. rec. if you have read the novel.
I loved Savannah, what a beautiful city. If you're looking for a cool/romantic b&b in the historic district, I liked Dresser-Palmer House. For activities - evening ghost tour with Savannah Walks, tour of Mercer-Williams House (read Midnight in the Garden of Good and Evil...great (true) story set in Savannah and this is the house where the main character lived), leave time to explore the different squares and lovely historic homes. We had a delicious dinner at Alligator Soul (upscale, romantic, cozy) and good lunches at Soho South Cafe and Vic's on the River (fried green tomato BLT). Those are my recommendations - unfortunately we didn't make it out to Tybee so I can't help you there.
I'm from Tybee. I'd suggest renting the kayak or paddle boards like the pp suggested, you can do a dolphin tour with Capt Mike, see the lighthouse/museum, Ft Pulaski, marine science center, go fishing with Capt Stan Marshland Adventures
Good places to eat:
Sundae Cafe- doesn't look like much, in a strip mall but it's fantastic AJs, Stingrays, Crab Shack- good for seafood, three different atmospheres Breakfast Club $$$ but delish, Sunrise- $ but good as well North Beach Grill- by the lighthouse
I am from Savannah and my parents have lived on Tybee for 20 years. We are actually heading there in a couple of weeks.
I love Fannie's on the Beach. It overooks the ocean, has live entertainment on the weekends, and has the most amazing jerk wings ever. Also Spanky's has the best chicken fingers! Both places are low-key/inexpensive. The lines are always out the door for the Breakfast Club, but I think it is way overpriced and not that great. We much prefer Sunrise (across the street on Butler.)
We have little kids, so we do kid-friendly stuff - but go into savannah and do a trolley tour, the pub crawls are fun too. Paula Deen's place is WAY overrated and crowded. We went to say we went, but were dissapointed. Uncle Bubba's (en route to Tybee) is also not that great. Get a drink and walk River Street in Savannah and take advantage of the open liquor laws (you can walk down the street with your drink.)
