I have nothing to add for route but if you have time in Knoxville prior to flying out Tupelo Honey is delicious. The only warning I would give is that the portions are HUGE so if there is a small size, get it.
I would take Path B, if only to avoid Knoxville traffic, which is heinous pretty much all the time. I go through Knoxville several times a year to visit family that lives on the other side and I would avoid Knoxville at all costs if I could. Depending on the time of day, traffic will be so bad.

I just consulted with a coworker who is familiar with both routes and she says option 1 is much better because the road around Jamestown is "busted up." She says stick to state highways and avoid county roads like the plague.
