How are you getting from the airport to Miami? How will you get from the hotel to the cruise terminal? You might want to check which hotels have free shuttles to the ship. I'll bet a bunch of them do.
This is probably one of those times I wouldn't do priceline. You have some pretty specific restrictions and I think that you want to know exactly where you're staying before you commit.

With flying into Ft Laurderdale and having two nights before your cruise, I would be renting a car. It shouldn't be very expensive for that short amount of time. IMO, Miami is best seen in a car. You could stay on South Beach and not leave the area for the entire time, and then take a cab to the cruise, but there is some nice stuff to see outside of South Beach. I prefer to have a car and stay somewhere nice and less expensive (using Priceline) in Coral Gables. Then I can do shops and restaurants in Coral Gables, see an attraction like Vizcaya Museum and Gardens, walk around the Biltmore Hotel, then spend a day at South Beach, etc. There should be rental car companies with a shuttle to the port, so on the day of your cruise you would just drop the car off and then take their shuttle to your cruise.
Ditto to dirtysouth's recs. We used to live in Miami & I used Priceline to get us a hotel in downtown Miami for one night pre-cruise next month. I ended up with the Marriott Biscayne Bay for $100/night & acc'g to the Marriott.com website immediately after I booked it via Priceline, it's a $309/ni room. You can PM me if you have specific questions although we haven't lived in Miami for almost 5 years now.
If you really don't want to rent a car, then stay in South Beach. You should be able to book Super Shuttle to take you from FLL airport to your hotel. I don't remember how much it will cost you since it's been a long time since I've had to do this, but it is doable. I get not wanting to drive and will not push you, but from a cost perspective, you will probably end up spending way more in cabs and shuttles than a rental car would be.
