Friday we did the amazing downtown art tour by the Detroit Experience Factory. Lunch at Slows in Corktown. DIA then dinner at Chartreuse. Saturday we went to the Detroit Institute of Bagels for breakfast and did a bus tour of breweries. Dinner was the only "disappointment" of the weekend. We went to Selden Standard. The food was great. The 2 hour wait for a table was less so (we should probably have just picked somewhere else, but were too beer-ed out to think that hard!). Sunday we went to Brooklyn Street Local and then walked along the river a bit. Stopped at Bon Bon Bon and then did a tour of the Fisher Building.
Book at Slows if you can. The art tour was fantastic and is definitely worth doing if you can. They do have some others as well. Stop at Bon Bon Bon.

I have a new restaurant to recommend! We had brunch at Bobcat Bonnie's in Corktown this Sunday and YUM. I had Captain Crunch encrusted French toast, H had some skillet thing called the Uncle Jesse, and we each had 3 drinks from the make-your-own Bloody Mary or mimosa bar, and the whole bill was under $40.

Also I've made the drive from Detroit to Chicago tons of times, and there's not a ton to do along the way except maybe Ann Arbor? (I assume you're taking I-94.) By the time you get to AA though you're almost in Detroit and might as well just keep going.

I'm sold. Their menu sounds delicious and you can't beat $3 Bloody Mary's.
The long weekend thread has most of my suggestions.
If you're looking for halfway points, Kalamazoo could be an option. There's a few breweries there (Bell's, Arcadia) and it's a small college town. Ditto a PP that Ann Arbor is the best midway point, but it's 30min from Detroit so maybe not worth stopping in when you'd be so close to your destination.
If you're into antiques at all, drive on 12 from Ann Arbor to Coldwater, then take 94 the rest of the way. On 12 you go through many little towns with many antique stores (Allen, MI even has a historical village with buildings brought in from surrounding counties). The drawback to this "scenic" route is that it's much slower than the interstate.
In the evening you can walk through Campus Martius park then down Woodward to the Riverwalk. It's lovely, and you can wave to Canada.
I second Roast, and also recommend fancy drinks at Coach Insignia, assume you're not afraid of heights and/or elevator rides that seemingly never end. I maybe had a panic attack in there the first time. I'm fine.

OMG. That elevator ride nearly did me in. I had to stare at my feet for the last half of the ride. I got nervous as our dinner ended knowing I had to ride it again! The views are worth it once you're up there!

I haven't either and I live less than a block away! Ha! I ate at the one in midtown and meh. (But I'm a bit of a bbq snob because my DH used to do competitions.)

The food at Wright & Co is terrific too. For drinks I like Two James or Gold Cash Gold. The lavender latte at Great Lakes is delicious, and the coffee at the bar inside Shinola is great too.
I'm easy to please but I love Andiamo's for Italian in the Ren Cen. And anything in Greektown. A Tigers game would be fun, it's a great ballpark - if you like baseball that is. Walk around Belle Isle too - it's pretty!
And I also recommend Campus Martius.
If you were bringing kids I would recommend the new hands on museum that the DNR opened along the river - but since you're not I'm putting it here for anyone else that might be interested! We took Morgan this past Spring and it was so fun!

We just went with some friends last month and I almost lost it too! I had to close my eyes and basically crouch down and old my husband for dear life-so embarrassing. Its like my two least favorite things in the world heights and elevators combined. Worth it for the view and food though in the end

I don't know if you're the gambling type, but there's a great casino in Battle Creek (right off 94, about 2 hours before you get to Detroit) called Firekeepers. The BBQ restaurant inside is really good; I'm not a foodie by any means, but we enjoyed it.
