Shaya is newish and fabulous. Not typical NOLA food, but I dream about it sometimes. I also love Domenica and August. For less fancy meals try District Donuts (not just for donuts), Rum House, Dat Dog, Company Burger, Surrey's, Ruby Slipper, Red Gravy. I could go on forever. We like to eat here.
I would try to get a cemetery tour through Save Our Cemeteries if you're into history at all. My biggest piece of advice is don't just stay in the FQ. Take a streetcar or Uber to the Garden District or Uptown and walk around. I live Uptown, so I'm biased, but it's lovely.
Oh, and if you want great grilled oysters, I would do Royal House Oyster Bar, Drago's, or Casamento's if they are open (only open during oyster season). Acme is not my favorite but it's fine if you want the experience.
Skip Pat o's hurricanes. The piano bar can be fun, but that drink sucks. There are so many places now that make a hurricane with real fruit juice that's delicious.
I like Muriel's on Jackson Square as well. Walk the royal street galleries, ride the streetcar into the garden district, cemetery tour, and eat.

I agree with these suggestions over Acme. Plus, you won't be stuck waiting in the ridiculous line for Acme.
I second Kirkette's recommendation about riding the St. Charles streetcar to the end of the line. You can see all the old homes and neighborhoods pass you by and it's so pretty! DH and I loved doing that when we were there for a couple days this past May.
Also, if you have interest in jazz, go to Preservation Hall one night. It's a very small venue, and if you don't get there super early before the performance, it'll be standing room only. But it's a very short performance (maybe half an hour, if I recall) and it's good, classic jazz.
Check out Mardis Gras World if you haven't already. It's a big warehouse where they make and store most of the Mardis Gras floats, and you can get a tour. It was interesting hearing the whole history of Mardis Gras and seeing the floats and props up close.
We also enjoyed the Aquarium of the Americas. Also, check out the French Market if you haven't been there yet.
And, of course, Cafe Du Monde! Cafe Beignet was also good, and had a nice atmosphere.
If you're there on a weekday, go to Commander's Palace for lunch. They have $0.25 martinis during lunch!!
Listen to jazz at Snug Harbor.
Other things: Cochon and/or Cochon Butcher Ruby Slipper for breakfast/brunch - so good we went there two days in a row Jacque Imo's Gator Tour (so fun) Drinks at the Carousel Bar

Cane and Table for brunch MoPho was not in the quarter area ( we were visiting friends who drove us there) but was a pretty tasty meal Drinks at Muriel's The Blue Nile for jazz (on Frenchman st) bonus if Kermit Ruffins is playing.
