Do you get the apple in the center at all or just the loading pinwheel? How long have you let it sit to see if it will progress further? How old is the machine?
My MacBook did this right before finals week my junior year, the hard drive was fried so it couldn't find the os files.

I have the apple and the pinwheel. It's about a year and a half old. I went and got dressed and it was still being an asshole. Then I did a start up disk repair, which said it was okay, restarted again and it's still being an asshole.
Back everything up to something like dropbox, email it to yourself, whatever. It could be that your harddrive is starting to fail or (very rare) you've picked up some kind of virus. I would find an authorized Apple repair place in your area and see if they can look it over.
It is most likely just the hard drive, most computer companies are notorious for putting in cheaper storage to keep their costs low. I had mine looked at and the HDD replaced in 2008 and I think I paid $250 for everything (re-installing all of the OS software, updates, etc.) and it has been fine since. I actually just gave it to my dad because I don't have much of a use for it anymore.
