As I mentioned in the Moloka`i post, the main activities are golf, exploring the town (takes no more than two hours, lol), exploring the island (takes half a day, maybe), horseback riding, and eating expensive food.
I think 3-4 days is enough, but it depends on how bored you get. You can also take the ferry to Lahaina for a day to break things up.
Both hotels are lovely. It just depends whether you prefer beach or mountain. I stayed at the lodge (Ko`ele) because it was cooler temperature-wise and seemed cozier, but spent a lot of time at the pool at Manele, too; there's a shuttle which will take you from one hotel to the other and stop in town in between.

Thanks. This is good to know. I was thinking 2 days, so it sounds like that would be plenty of time to see everything. Are there restaurants in the town area or do you have to eat at the hotel?

I wrote them at the same time and thought you would be interested. It's kind of funny that both islands came up on the same day
There are restaurants in town, though not all of them stay open for dinner. But there are definitely options for breakfast and lunch that would be cheaper than the hotels.
