I would say a minimum of a couple of hours but you could probably do longer depending on your level of interest with history/architecture, etc. I think that many people come to Cordoba solely for the Mezquita but its Juderia (old Jewish quarter) is a fantastic place to wander aimlessly along its narrow medieval streets and a lot of people miss out on that. I'm not sure what day of the week you'd be going but if it's a weekend try to get there first thing as the crowds for the Mezquita can be crazy. Cordoba has only one of three (I think that's the number) synagogues left that pre-date the destruction of the Inquisition. Small, but fascinating. We also had lunch at a fantastic Moroccan place. I'm more partial to Sevilla but the Mezquita is just one of those places you'll never forget and be utterly amazed by.
I think a long day trip is fine. Plan to arrive in the morning and leave by 5 or 6. That will give you time to get back to Seville, rest and be ready to go out for dinner around 10.
We stayed 3 nights and weren't bored at all so really, as long as you can without making a crazy day. The Mezquita is amazing and we just really liked the city. I agree with Anna - arrive in the morning, see something, bit of a leisurely lunch, wander around a bit, see something else, head back would work well.
