A job that is equal to my supervisor's position is coming open in another office close to us.
I'm toying with the idea of applying for it. I know that I could do it, and do it well. But I think it requires a Master's degree.
EDIT: Oh, and a follow up question- I think there is a good possibility that if I apply, my supervisor will find out. Any thoughts on that?
Also, my OTHER supervisor (not my direct supervisor)- should I talk to her about it? I think that she feels my pain in regards to my direct supervisor and might have some ideas.
Go for it. If they think you are not qualified they won't call you in for an interview. No point in you making that decision for them before they've had a chance to see your qualifications/expertise.
Definitely go for it--so many times jobs post crazy requirements, kind of like a dream list of qualifications, but they're often willing to consider people who don't meet every single requirement. Anyway, it can't hurt to apply!

And you said it's for the company you're already working for, right? That's actually good, because your employers will see that you're motivated to move up, and even if they decide on someone else for that position it might open doors in the future.
I just got an in-company promotion/move to a different department and I didn't have all of the "requirements". The list of theirs was a dream list.
ETA: about your suprevisor - is your company big on internal advancement? my company is, so when you apply internally, it is an open book (well, to you, your supervisor and the hiring manager) and no back lash.
Apply! In your cover letter explain that while you are not currently a supervisor for X you have been doing X job for X years and are well versed in the ins and outs of it and are looking for an opportunity to advance.
