Same company? I would say yes either way but ABSOLUTELY if it is internal and you want to stay in your company. Even if they don't select you for this, you might be perfect for the next opportunity and remember your resume.
Yes. You have no way of knowing what the rest of the applicant pool looks like; they might not get anyone who does have exactly what they want, and you might bring a skill set they didn't know or didn't post that they needed. Part of what got me my last IT job was my training experience - it wasn't anywhere on the job posting but they desperately needed that skill in the department.

This. When I applied for my current position(it was internal), I was missing one of their qualifications. My boss was very honest that they view it as a general outline but If they found the right candidate who was missing a qualification they would still get the job.
