Is there someone in HR you can speak to? Cause her lying is not cool and normally, I'd say confront head on, but it's more complicated with your friend in the middle now.
It seems like your manager doesn't want to let you out of your position, and yet, you probably are ready for a promotion for a while. Can you speak to her manager?

And fwiw, I think its a huge line of crap (not that you aren't good at what you do) when a manager says that they can't promote you because you're too good at your job. A good manager encourages an employee to grow and further their careers even if it means having to hire someone new. Don't be punished by your bad/lazy manager for doing a good job.

When was the last time you discussed career growth with your manager? I would make sure to bring it up soon and maybe say that you feel like you've been stagnating and while you like it there (do you?) it's gotten to the point you might start looking for career growth opportunities elsewhere.

Also is there someone you can recommend who you think would be a good admin? That might make it easier for them to transfer/promote you if they know there's a reliable replacement for you.
Agree w/ all pp. This is bs and you should look for something else. But, if you want to stay or are worried about finding something in this economy, I would say something, like "I know that ____ is leaving and I don't know if you have any plans for _____'s position, but I wanted to let you know that I am interested." And maybe, "Please let me know when we can discuss it and/or what the process is to apply."
